Negative earth - shouldn't have needed to check really. With a line fuse in the main positive cable. As has been said, it should run quite happily without a battery. If it won't start a quick trawl of the forum will reveal the usual ignition suspects :wink:

I'll go and have a look later to confirm the polarity, but as mine has a 12v conversion (as well as a 350 engine) the battery isn't standard.

Is there a dedicated earth connection from the engine to frame or are you just relying on the engine mountings? I have a cable running from the crankcase screw that is just behind the lug for the clutch cable to a common earth point on the frame for the ignition and charging system. That's with a 35...

I've never disconnected the battery leads on my 375/2C other than to change the battery in 30 years. I can't see a proper trickle charger being a problem and I regularly used an Optimate and now a cheap Aldi 'smart charger'.

I had to do the front wheel bearings on mine last year for the MoT and I know they had never been changed before in 30 years so was dreading it. No problem though once I had heated things up. As you have a multimeter, all you may need is the recommended resistances and voltages measured at the regulator/rectifier connections and the stator coils. That may be in the workshop manual if you can get hold of one - possibly on-line.
